Review[Recent research advances on cryopreserved platelets].
The right choice of frozen protective agents and additives is an important factor to ensure the quality of frozen platelets. The immediate hemostatic function of frozen platelet in vivo is superior to liquid-stored platelets. In order to ensure the quality of frozen platelets better, it is important to understand the role of dimethylsulfoxide (DMSO) in the preservation of frozen platelets and its mechanism, and to understand the mechanism of DMSO enhancing the hemostatic function of frozen platelets and the effects of different factors on the frozen platelets. In this review, the long-term preservation of frozen platelets and its quality standards, the mechanism of DMSO effect on the molecular changes and inhibition of frozen platelets, different factors influencing preservation freezing platelets and the test of preservation effects, the application of frozen platelets to the military operation and disaster relief, the canine frozen platelet studies and so on are summarized.

[Effects of transcription factor T-bet, GATA-3, FoxP3 and CD4(+)CD25(+)regulatory T cells in pathogenesis of child Hench-Schonlein purpura].
The aim of this study was to investigate the effects of transcription factors T-bet, GATA-3 in the pathogenesis of Hench-Schonlein purpura (HSP) in children, the relationship between CD4(+)CD25(+)regulatory T cells, transcription factor FoxP3 and the development of child HSP, and the molecular mechanisms of Th1/Th2 imbalance of child HSP at acute phase, so as to may provide a new approach and strategy for the treatment of HSP at the molecular levels. The expression of T-bet, GATA-3 and FoxP3 mRNA were detected by real time PCR using SYBR Green I in 46 patients with HSP at acute phase and 30 healthy children as controls. The expression of T lymphocyte subsets CD4(+)CD25(+) in peripheral blood mononuclear cells was detected by flow cytometry. ⋯ At acute phase of child HSP, the expression of CD4(+)CD25(+)Treg and its special transcription factor FoxP3 mRNA are down-regulated. Treg cells decreases, which indicates that insufficient immunosuppressive effects resulting from the reduction of Treg cells may be one of the important reason in the immune imbalance of HSP acute phase. This study provides experimental evidence for illustrating the pathogenesis of HSP from the molecular mechanism of Treg cells and its regulation, and also provides a new thinking and new strategies for the treatment of HSP at molecular levels.

Review[Advancement of peptide vaccines for hematologic malignancies].
Despite recent significant advances in the treatment of hematological malignancies, relapse of this disease is of great note with the existence of the minimal residual disease (MRD). Tumour peptide vaccine seems to be one of the effective immunotherapies for eliminating tumor cells of MRD. This review focuses on the late results of clinical trails of peptide vaccination protocols targeting WT1, RHAMM, BCR-ABL, PR1 in hematological malignancies and the development of specific immune responses to PRAME and Survivin peptides. An outlook to heteroclitic peptides, new adjuvants, combined peptide vaccines and Ad-tWT1 vaccine is also given to further explore the possibility to enhance the efficacy of the peptide vaccine.

[Influence of BCR-ABL inhibitor STI571 on SARI expression in K562 cells].
In order to investigate the molecular mechanisms of SARI expression regulation in chronic myeloid leukemia (CML), 46 patients with CML and 40 healthy volunteers were recruited in this study. SARI expression in the peripheral blood mononuclear cells (PBMNC) of CML patients and healthy volunteers was assayed by using real-time quantitative PCR. K562 cells were in vitro incubated with the BCR-ABL inhibitor STI571 (imatinib) at 37°C and 5% CO2 for 24 hours, then SARI expression was detected by using real-time quantitative PCR. ⋯ After exposure of K562 cells to STI571 (2.5 µmol/L) for 24 hours, the SARI expression was higher than that in K562 cells treated without STI571 (p < 0.001). It is concluded that the suppression of SARI expression is involved in CML pathogenesis, and BCR-ABL mediates the down-regulation of SARI mRNA expression in K562 cells. These findings suggest a new orientation for gene therapy in CML patients.

[Immunophenotype characteristics and prognosis of acute leukemia patients with cross expressing lymphoid and myeloid lineage associated antigens].
The aim of study was to investigate the immunophenotype characteristics and prognosis of acute leukemia patients with cross-expressing lymphoid and myeloid lineage-associated antigens. The immunophenotypes of leukemic cells were examined by using flow cytometry. All patients were classified into several groups according to FAB subtypes and immunophenotyping. ⋯ CR ratio and RFS of CD13(+) ALL and CD13/CD33(+) ALL patients had no significant difference when compared with My(-) ALL patients. It is concluded that the importance of cross-expressing antigens for prognosis of patients should be analyzed concretely. CD56(+) AML and Ly ≥ 2(+) AML have bad prognosis, while other cross-expressed lymphoid and myeloid lineage-associated antigens have no impact on prognosis of acute leukemia patients.
