Review[Research Progress on Treating Acute Myeloid Leukemia by Midostaurin].
FLT3 gene mutations occurred in approximately 30% of acute myeloid leukemia (AML) patients, which is closely associated with the occurrence, development and poor prognosis of AML. The therapy targeting at FLT3 mutations might be a promising treatment for AML. Midostaurin can inhibit the activities of III receptor tyrosine kinase encoded by FLT3 gene, induce cell cycle arrest and has a apoptotic effect on primitive AML cells of FLT3 -mutant, FLT3 wild-type and the expression of FLT3 mutated receptor. In view of this, the association between FLT3 mutations and AML, and research advances and clinical applications of midostaurin on the treatment of AML especially for FLT3 mutated AML, are reviewed.

Review[Treatment of lymphoblastic leukemia with CD19-specific modified chimeric antigen receptor T cells].
Bioengineered T cells, which are the genetically manipulated T cells to express chimeric antigen receptor T Cell (CAR T) against leukemia-associated specific antigens, were applied to treat acute and chronic lymphocytic leukemia with CAR T. CAR T cells combined with cell-surface binding site and anti-CD19 chimeric antigen receptor can treat diseases through T cells transfection. ⋯ CAR T cells can proliferate by 1000 times and differentiate in vivo by the CD19 antigen stimulation, therefore, kill the acute and chronic lymphocytic leukemia cells effectively. This article briefly reviews the CAR T cells and the effect of CAR T cells on acute and chronic lymphoblastic leukemia.

Review[CAL-101,a novel agent of targeted therapy in hematological malignancies].
CAL-101 is a selective inhibitor of the phosphatidylinositol-3 kinase (PI3K), it inhibits the survival, proliferation and migration of tumor cells by directly inducing apoptosis and inhibiting micro-environmental interactions. It has been determined that the P110δ isoforms of PI3K expressed primarily in cells of hematopoietic lineage, such as B and T cells. This review focuses on the target, mechanism of action, the use and prospect of CAL-101 in tumors of blood and lymph systems.

Review[Advances in immunotherapy of hematological malignancies by using chimeric antigen receptor-modified lymphocytes].
Chimeric antigen receptors (CAR) are fusion proteins between single-chain variable fragments (scFv) from monoclonal antibodies and signaling domains of T-cells, which allow T-cells recognize specific cell-surface targets in an MHC-unrestricted fashion. The structure of CAR has changed over time, from first generation CAR (scFv + signaling moiety) to 2 and 3 generation CAR (combined with one or multiple costimulatory endodomains, such as CD28, 4-1BB and OX40), which enhance persistence, expansion and cytotoxicity of CAR. Many clinical trials treating hematological malignancies using the CAR-modified T-cells targeting CD19 and CD20 are under evaluation or even finished. ⋯ As to the treatment toxicity, CAR modified T-cell infusion is tolerant and safe in most patients. However, insertional mutagenesis, off-target effect and inflammatory response are safety issues surrounding CAR-modified T-cell therapy. In this review, the use of CAR technique in treatment of hematologic malignancies and evaluation of CAR safety are summarized.

Review[Recent research advances on cryopreserved platelets].
The right choice of frozen protective agents and additives is an important factor to ensure the quality of frozen platelets. The immediate hemostatic function of frozen platelet in vivo is superior to liquid-stored platelets. In order to ensure the quality of frozen platelets better, it is important to understand the role of dimethylsulfoxide (DMSO) in the preservation of frozen platelets and its mechanism, and to understand the mechanism of DMSO enhancing the hemostatic function of frozen platelets and the effects of different factors on the frozen platelets. In this review, the long-term preservation of frozen platelets and its quality standards, the mechanism of DMSO effect on the molecular changes and inhibition of frozen platelets, different factors influencing preservation freezing platelets and the test of preservation effects, the application of frozen platelets to the military operation and disaster relief, the canine frozen platelet studies and so on are summarized.
