An open labeled randomized controlled trial of pregabalin versus amitriptyline in chronic low backache.
There is no head on comparison of amitriptyline (AMT) and pregabalin (PG) in relieving pain and disability in chronic low backache (CLBA). This randomized controlled trial reports the efficacy and safety of AMT and PG in CLBA. ⋯ AMT and PG are effective in CLBA but AMT reduced pain and disability significantly compared to PG.

Randomized Controlled Trial Comparative StudyEquivalency of tricyclic antidepressants in open-label neuropathic pain study.
To compare adverse effects, tolerability and efficacy of the tricyclic antidepressants (TCAs) amitriptyline and nortriptyline in management of neuropathic pain due to peripheral neuropathy (PN). ⋯ Amitriptyline and nortriptyline are equivalent for overall adverse effects and discontinuation rates. Either TCA should be equally considered for use in neuropathic pain due to PN. When used as monotherapy or as part of adjuvant therapy, either TCA can be expected to provide approximately 23-26% visual analog scale pain reduction if tolerated. Discontinuations due to inefficacy or adverse effects can be anticipated in 26-37% of patients initiated on either TCA for PN-associated neuropathic pain.

Cognitive behavioral therapy plus amitriptyline for chronic migraine in children and adolescents: a randomized clinical trial.
Early, safe, effective, and durable evidence-based interventions for children and adolescents with chronic migraine do not exist. ⋯ Among young persons with chronic migraine, the use of CBT plus amitriptyline resulted in greater reductions in days with headache and migraine-related disability compared with use of headache education plus amitriptyline. These findings support the efficacy of CBT in the treatment of chronic migraine in children and adolescents.

This study compares efficacy and safety of divalproate extended release (DVA-ER) and amitriptyline (AMT) in migraine. ⋯ Divalproate extended release is more effective at 3 months than AMT; however, at 6 months, both are equally effective in migraine prophylaxis.
