Broad bean (Vicia faba L.), a common vegetable, belongs to the family Fabaceae and is consumed worldwide. Limited studies have been done on allergenicity of broad beans. The aim of this study was to determine if broad bean proteins have the ability to elicit allergic responses due to the presence of clinically relevant allergenic proteins. ⋯ This study inferred that broad bean proteins have the ability to elicit allergic responses due to the presence of clinically relevant allergenic proteins.

To evaluate the benefits and risks of pitolisant (a wake-enhancing drug that increases the histamine release in the brain by blocking presynaptic H3 histamine reuptake) in patients with idiopathic (IH) and symptomatic (SH) hypersomnia plus sleepiness refractory to available stimulants (modafinil, methylphenidate, mazindol, sodium oxybate, and d-amphetamine). ⋯ Pitolisant had a long-term favorable benefit/risk ratio in 23-38% of drug-resistant patients with IH and SH, suggesting that histamine neurons can be stimulated in severe idiopathic and symptomatic hypersomnia.

Myelinated Ah-type trigeminal ganglion neurons in female rats: neuroexcitability, chemosensitivity to histamine, and potential clinical impact.
Migraine is a chronic neurological disorder characterized by recurrent moderate-to-severe headaches often associated with numerous autonomic nervous system symptoms, and it is more prevalent in women. To fully understand the underlying mechanism, standard electrophysiology was performed with trigeminal ganglion neurons (TGNs) isolated from adult rats of both genders using the whole-cell patch clamp technique to test the distribution, neuroexcitability, and chemosensitivity to histamine. In addition to traditionally classified A- and C-type TGNs, myelinated Ah-type TGNs were also observed in females. ⋯ Iberiotoxin and NS11021 significantly altered the discharge profiles of Ah-type TGNs. Finally, Ah-type TGNs showed a more potent reaction to histamine, with relatively larger inward currents and membrane depolarization compared with C-types. These data provide evidence of the gender-specific distribution of myelinated Ah-type TGNs in adult female rats, characterized by a low threshold and high frequency of firing that are at least partially attributable to persistent TTX-R Na(+) and BK-KCa channel expression and potent chemosensitivity to histamine, suggesting that Ah-type TGNs may play a key role in gender differences in migraine.

Anaesthetic hypersensitivity reactions can be IgE- or not IgE-mediated and are a challenge to find the causal agent. Histamine and tryptase determination are classically considered useful in the diagnosis of these reactions. The aim of our study was to assess the diagnostic usefulness of plasma histamine and different cut-off points of serum tryptase. ⋯ The best criterion for discriminating IgE- and non IgE-mediated hypersensitivity reactions in anaesthesia was a tryptase value exceeding twice the basal one.

Astrocytic glutamate transporter 1 (GLT-1) is responsible for the majority of extracellular glutamate clearance and is essential for preventing excitotoxicity in the brain. Up-regulation of GLT-1 shows benefit effect on ischemia-induced neuronal damage. In present study, we examined the effect of histamine, a neurotransmitter or neuromodulator, on GLT-1 expression and function. ⋯ In addition, histidine, the precursor of histamine, also showed neuroprotection against ischemic injury, which was accompanied by reversion of declined expression of GLT-1 in adult rats subjected to middle cerebral artery occlusion (MCAO). These neuroprotective effects of histamine/histidine were blocked by GLT-1 specific inhibitor dihydrokainate or H1 receptor antagonist. In summary, our results suggest that histamine up-regulates GLT-1 expression and function via astrocytic H1 receptors, thus resulting in neuroprotection against excitotoxicity and ischemic injury.
